Area of Application
Poly-HRP products are supermolecular enzyme complexes that contain up to 400 enzyme molecules and can be conjugated with various ligands and receptors, such as those located on antibodies, antigens, streptavidin, and DNA. These conjugates can be widely used in assay systems such as ELISAs, immunoblotting, immunofiltration, or DNA/RNA hybridization. Their specific activity is superior to conventional conjugates by several orders of magnitude . They allow an amelioration of assay sensitivity, shorter assay times, and limit reagent consumption. Particularly suited to microstructured chips and rapid tests, poly-HRP products are widely used to optimize conventional ELISA tests.

PolyHRP Structure and Composition
To create a PolyHRP product, the enzyme molecules are cross linked using bifunctional crosslinkers with oligomer complexes consisting of 20, 40, or 80 molecules. The next step involves joining five of these sub-units – called “Homoblockpolymeres”– together to form a supermolecular complex. The complex can furthermore be conjugated with other biologically active molecules, for example antibodies.
Senova currently offers three standard sizes of Poly HRP: PolyHRP20, PolyHRP40, PolyHRP80. PolyHRP can be conjugated with a wide number of ligands and receptors.

Rapid Tests Based on PolyHRP
Poly HRP conjugates are used for the production of rapid immuno assays with microcolumns. These assays are based on the 3D immunofiltration principle and allow a quantitative determination of analytes within a brief timeframe of 10 to 30 minutes. By using PolyHRP conjugates, results similar to those yielded by ultra sensitive ELISAs in the range of less than 1pg/ml can be obtained.
